Exposure of the nonsteroidal anti-inflammatory drug suprofen (SUP) to UV-radiation results in the formation of radicals, reactive oxygen species (ROS), photodecarboxylated products and photoadducts with biomacromolecules. Using an ex vivo pigskin explant model, we investigated whether topical coapplication of the water-soluble antioxidants vitamin C (Lascorbic acid, ASC), N-acetyl-L-cysteine (NAC) or L-cysteine ethylester (CYSET) with SUP reduced ultraviolet A (UVA)-induced decomposition of SUP. UVA-induced changes in antioxidant bioavailability in the stratum corneum and epidermis were also studied. Epidermal bioavailability of SUP in sham-irradiated pigskin increased 2.2- to 4.1-fold after the lowest antioxidant doses (P < 0.05). As compared with no applied antioxidant, increasing doses of all tested antioxidants resulted in increased levels of SUP and decreased levels of photoproducts (P < 0.05). A maximal protection against SUP photodegradation of 70% was found after an ASC dose of 1 micromol/cm2; these values were 60% for a NAC dose of 10 micromol/cm2 and 50% for a CYSET dose of 5 micromol/cm2. Skin antioxidant levels increased with increasing applied dose (P < 0.05); the bioavailability of CYSET was approximately three-fold lower than that of ASC and NAC. UVA exposure resulted in 30-50% consumption of the topically applied ASC or NAC in the stratum corneum, whereas CYSET was not consumed. In conclusion, the topically applied water-soluble antioxidants ASC, NAC and CYSET protect against UVA-induced decomposition of SUP by scavenging radicals and ROS. Coapplication of these antioxidants may therefore be an effective way to reduce or prevent the phototoxic effects of SUP in vivo.  相似文献

We review the recent progress in the modeling of plasmas or ionized gases, with compositions compatible with that of primordial atmospheres. The plasma kinetics involves elementary processes by which free electrons ultimately activate weakly reactive molecules, such as carbon dioxide or methane, thereby potentially starting prebiotic reaction chains. These processes include electron–molecule reactions and energy exchanges between molecules. They are basic processes, for example, in the famous Miller-Urey experiment, and become relevant in any prebiotic scenario where the primordial atmosphere is significantly ionized by electrical activity, photoionization or meteor phenomena. The kinetics of plasma displays remarkable complexity due to the non-equilibrium features of the energy distributions involved. In particular, we argue that two concepts developed by the plasma modeling community, the electron velocity distribution function and the vibrational distribution function, may unlock much new information and provide insight into prebiotic processes initiated by electron–molecule collisions.  相似文献

Triorganotin(IV) derivatives containing the anionic ligand bis(1‐methyl‐1H‐imidazol‐2‐ylthio)acetate [(S‐tim)2CHCO2] were synthesized from the reaction between R3SnCl acceptors (R = Me and Ph) and the sodium salt of the ligand. Mono‐nuclear complexes of the type [(S‐tim)2CHCO2]SnR3 were obtained, which were fully characterized by elemental analyses and FT‐IR in the solid state, and by NMR (1H, 13C and 119Sn) spectroscopy and electrospray ionization mass in solution. The toxic effects shown by these compounds on trout erythrocyte components showed that the toxicity of the organotin(IV) complexes depends on the nature and on the lipophilicity of the substituents on the metal centre. The nature of the interaction of isocyanic acid (HNCO) with the active centers at the ideal anatase TiO2 (1 0 1) surface were studied using ab initio density functional theory (DFT) method with a cluster model. Two types of adsorption of isocyanic acid are found to be likely at (1 0 1) surface – dissociative and molecular adsorption. Only molecular adsorption of HNCO leads to the direct weakening and further splitting of the NC bond, which is a necessary step for the hydrolysis of isocyanic acid. During molecular adsorption of HNCO, an energetically stable intermediate surface complex is created with an adsorption energy of −1.33 eV, in which the HNCO skeleton is changing due to new strong bonds between C–Os and N–Tis. Based on the existence of this intermediate complex a probable reaction pathway for the hydrolysis of HNCO over the ideal anatase (1 0 1) surface was developed. A surface oxygen vacancy was formed after the decomposition of the intermediate complex and CO2 desorption. Afterwards, water adsorbs at the oxygen vacancy site and NH3 is successively formed. The HNCO hydrolysis over TiO2 was found to be energetically favorable with global energy gain of about −2.08 eV.  相似文献

We address an optimization problem in which two agents, each with a set of weighted items, compete in order to maximize the total weight of their winning sets. The latter are built according to a sequential game consisting in a fixed number of rounds. In every round each agent submits one item for possible inclusion in its winning set. We study two natural rules to decide the winner of each round.For both rules we deal with the problem from different perspectives. From a centralized point of view, we investigate (i) the structure and the number of efficient (i.e. Pareto optimal) solutions, (ii) the complexity of finding such solutions, (iii) the best-worst ratio, i.e. the ratio between the efficient solution with largest and smallest total weight, and (iv) existence of Nash equilibria.Finally, we address the problem from a single agent perspective. We consider preventive or maximin strategies, optimizing the objective of the agent in the worst case, and best response strategies, where the items submitted by the other agent are known in advance either in each round (on-line) or for the whole game (off-line).  相似文献
